Trace Lines in 3-band image with varying hues

Hi! Can someone help me figure out the best way to take images (I have many variations) like this:

and annotate them like this (where the blue and yellow lines are data whose locations in the array can be stored)?

So far, I have tried using various combinations of imbinarize,imadjust,imdiffusefilt and edge in addition to trying to manually find RGB values that will separate the different segments. The lines I am trying to annotate vary in length and position.
