Detecting nuclei from a microscopic image

I have these microscopic images from which I need to detect the nuclei. I sharpened the image using "localcontrast()" and the nuclei look pretty decent. After, I tried reducing noise using medfilt3() but most of the nuclei disappeared. How can I solve this problem?
