Generally speaking, Yes. This is usually done for "gesture recognition".
There can be challenges to get it to work well at different scales (in 3d) in cluttered environments with different skin tones and other objects in view that might match skin tone by accident. If you add in mitts or gloves it can also get more difficult.

Thank you for answer, i need Human hand detection using Normal RGB camera , dont use human skin color, dont use template , no use XML , only shape based to detect the human hand , can you help me sir ?
No, under those restrictions I have no assistance to offer.
Thank you for your reply , sir, i don't need your assistance, can you tell some idea ?
When I say I have no assistance I include ideas about how you could proceed. You have ruled out enough possibilities that I am not sure that it is possible to do what you want to do. You will need to research algorithms through a site such as http://scholar.google.com and you might find a solution among the hundreds of thousands of citations.
