modify the audio gain of a specific frequency

I'm not very expert on audio manipolation ad i want create a script that can modify the gain of a specific frequency ( i have seen that it should be use a filter but i didn't understand how create it and how use it)

firls function of Signal Processing Toolbox can be used to create filters in matlab.
